Directions for use
The lyophilized preparation can be stored at 2~8 °C. If aliquots are required, they can be dissolved with water for injection, normal saline, culture medium or PBS, dissolved into small aliquots, and stored at -20°C for 6 months and -80°C for 12 months, with a diluted concentration of not less than 100μg/mL. Avoid repeated freeze-thaw cycles.
Activated T cells generally do not survive long-term in vitro culture, but the addition of IL-2 allows them to proliferate for a long time. Quiescent T cells do not express IL-2R on the surface and do not respond to IL-2; T cells can only express IL-2R after activation by mitogen or other stimuli, and become target cells of IL-2; IL-2 in turn can induce increased IL-2R expression in target cells. The expression of IL-2R on T cells is one-time, generally peaking 2~3 days after activation and disappearing around 6~10 days. As IL-2R disappears, T cells lose their ability to respond to IL-2. Therefore, in order to maintain the long-term growth of normal T cells in vitro, it is necessary to continuously stimulate T cells with mitogen or other stimuli to maintain IL-2R expression. NK cells are the only lymphoid cells that normally express IL-2R and therefore remain reactive to IL-2 at all times. However, quiescent NK cells express only the β and γ chains of IL-2R, which have a low affinity for IL-2 and can only respond to high concentrations of IL-2. Once NK cells are activated, they express the α chain of IL-2R, which acts as a high-affinity receptor. Suitable for the production of cell therapy products.
